When a custom colony has been set in place, is it possible to move it elsewhere, later on?

No, your colony will be there forever. Well, technically you can through save editing, but that's not something you can do on a switch.
If you're really unsure about the spot, you can copy your save (it's one of the options when you go to load a file) right before you make a colony so you have a chance to go back if you change your mind. However, you would of course lose any progress you made after making the colony if you decided to switch.
